Ces cellules offrent la possibilit\u00e9 de g\u00e9n\u00e9rer \u00e0 volont\u00e9 des cellules humaines de diff\u00e9rents types, constituant ainsi des mod\u00e8les de recherche uniques pour la compr\u00e9hension des pathologies et l\u2019\u00e9laboration de nouvelles approches th\u00e9rapeutiques.<\/em><\/p>\n\n\n\n<div class=\"wp-block-columns is-layout-flex wp-container-core-columns-is-layout-28f84493 wp-block-columns-is-layout-flex\">\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\">\n<h3 class=\"wp-block-heading\" id=\"h-axes-de-recherche\" style=\"text-transform:none\">Axes de Recherche : <\/h3>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-maladies-neurodegeneratives-et-organoides-cerebraux\"><strong>Maladies neurod\u00e9g\u00e9n\u00e9ratives et organo\u00efdes c\u00e9r\u00e9braux<\/strong><\/h4>\n\n\n\n<p>Nous d\u00e9veloppons de nouveaux mod\u00e8les in vitro d\u00e9riv\u00e9s de cellules iPS pour l\u2019\u00e9tude de pathologies telles que la maladie d\u2019Alzheimer ou les maladies \u00e0 prions. Gr\u00e2ce \u00e0 leurs propri\u00e9t\u00e9s d\u2019auto-organisation, ces cellules permettent \u00e9galement de g\u00e9n\u00e9rer des organo\u00efdes c\u00e9r\u00e9braux (\u201cmini-cerveaux\u201d), qui reproduisent certaines caract\u00e9ristiques du tissu humain. Ces mod\u00e8les repr\u00e9sentent une alternative prometteuse aux approches traditionnelles et ouvrent la voie \u00e0 des outils de criblage pharmacologique plus fiables pour la recherche acad\u00e9mique et l\u2019industrie pharmaceutique.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-ingenierie-de-la-pluripotence\">Ing\u00e9nierie de la Pluripotence<\/h4>\n\n\n\n<p>Nous mettons au point des techniques innovantes de reprogrammation et de diff\u00e9renciation cellulaire afin d\u2019\u00e9largir la palette des mod\u00e8les disponibles. L\u2019\u00e9quipe d\u00e9veloppe et conserve des lign\u00e9es cellulaires bien caract\u00e9ris\u00e9es (Master Cell Bank), avec une attention particuli\u00e8re port\u00e9e \u00e0 la standardisation, au contr\u00f4le qualit\u00e9 et \u00e0 l\u2019int\u00e9gration de nouvelles technologies comme l\u2019imagerie avanc\u00e9e et l\u2019automatisation des cultures.<br>Par ailleurs, nous explorons la modification g\u00e9n\u00e9tique des cellules par l\u2019utilisation de vecteurs viraux ou par l\u2019\u00e9dition g\u00e9nomique via <strong>CRISPR-Cas9<\/strong>. Ces approches permettent de g\u00e9n\u00e9rer des mod\u00e8les cellulaires innovants, adapt\u00e9s \u00e0 l\u2019\u00e9tude de mutations sp\u00e9cifiques et au d\u00e9veloppement de nouvelles strat\u00e9gies th\u00e9rapeutiques.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-biologie-spatiale\">Biologie spatiale <\/h4>\n\n\n\n<p>Nous explorons le potentiel unique de la culture cellulaire en apesanteur. L\u2019\u00e9tude des effets de la microgravit\u00e9 sur des tissus c\u00e9r\u00e9braux humains in vitro constitue une approche innovante pour mieux comprendre la physiologie cellulaire et les m\u00e9canismes du vieillissement. Une premi\u00e8re exp\u00e9rience r\u00e9ussie a \u00e9t\u00e9 men\u00e9e dans le cadre du projet <strong>Cerebral Ageing<\/strong>, en collaboration avec l\u2019Institut Pasteur et le CNES.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-microfluidique\">Microfluidique<\/h4>\n\n\n\n<p>En collaboration avec des entreprises sp\u00e9cialis\u00e9es comme <strong>NETRI<\/strong>, nous explorons l\u2019utilisation des technologies microfluidiques pour accro\u00eetre la robustesse et la reproductibilit\u00e9 de nos mod\u00e8les. Ces syst\u00e8mes miniaturis\u00e9s permettent d\u2019automatiser certaines \u00e9tapes de culture et de cr\u00e9er des environnements plus proches de la physiologie, ouvrant ainsi la voie \u00e0 des applications plus accessibles \u00e0 l\u2019industrie.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-imagerie\">Imagerie <\/h4>\n\n\n\n<p>L\u2019imagerie, et en particulier l\u2019imagerie 3D, est un outil indispensable pour nos recherches. Elle repose sur des technologies vari\u00e9es et complexes qui permettent d\u2019analyser la structure et le fonctionnement des mod\u00e8les cellulaires et organo\u00efdes. D\u00e9couvertes \u00e0 partir d\u2019embryons humains en 1998, elles ont connu une r\u00e9volution en 2007 lorsque le Pr Yamanaka a d\u00e9montr\u00e9 qu\u2019il \u00e9tait possible de reprogrammer des cellules adultes en cellules souches pluripotentes induites (<strong>iPS<\/strong>), une avanc\u00e9e couronn\u00e9e par le prix Nobel en 2012.<\/p>\n\n\n\n<p class=\"has-tertiary-background-color has-background\"><\/p>\n\n\n\n<h3 class=\"wp-block-heading has-tertiary-color has-text-color\" id=\"h-que-sont-les-organoides-cerebraux\" style=\"padding-top:var(--wp--preset--spacing--30);padding-right:var(--wp--preset--spacing--30);padding-bottom:0;padding-left:var(--wp--preset--spacing--30)\">Que sont les organo\u00efdes c\u00e9r\u00e9braux ?<\/h3>\n\n\n\n<p><\/p>\n\n\n\n<p class=\"has-tertiary-color has-text-color has-small-font-size\" style=\"text-align: justify; hyphens: auto; line-height: 1.6; padding: 0 var(--wp--preset--spacing--30) var(--wp--preset--spacing--30) var(--wp--preset--spacing--30);\">Les organo\u00efdes c\u00e9r\u00e9braux sont des structures tridimensionnelles obtenues \u00e0 partir de cellules souches pluripotentes, d\u00e9crits pour la premi\u00e8re fois par Dr M Lancaster en 2013. Gr\u00e2ce \u00e0 leurs propri\u00e9t\u00e9s d\u2019auto-organisation, ces cellules forment des \u00ab mini-cerveaux \u00bb qui reproduisent certains aspects du d\u00e9veloppement et de l\u2019architecture du tissu c\u00e9r\u00e9bral humain.
