Ces ph\u00e9nom\u00e8nes sont centraux dans de nombreux domaines de la biotechnologie : sant\u00e9 (croissance tumorale et pathog\u00e8nes), agroalimentaire, cosm\u00e9tique, pharmaceutique ou encore bio\u00e9nergie, pour la production de mol\u00e9cules \u00e0 haute valeur ajout\u00e9e.<\/em><\/p>\n\n\n\n<div class=\"wp-block-columns is-layout-flex wp-container-core-columns-is-layout-ef06f3fc wp-block-columns-is-layout-flex\">\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\" style=\"flex-basis:66.66%\">\n<h3 class=\"wp-block-heading\" id=\"h-axes-de-recherche\">Axes de recherche<\/h3>\n\n\n\n<h5 class=\"wp-block-heading\" id=\"h-recherche-therapeutique-en-silico\"><strong>Recherche th\u00e9rapeutique en silico<\/strong><\/h5>\n\n\n\n<p>Cet axe vise \u00e0 identifier de nouvelles mol\u00e9cules ou combinaisons de m\u00e9dicaments capables d\u2019inhiber la prolif\u00e9ration cellulaire. Le BIRL s\u2019int\u00e9resse particuli\u00e8rement au transfert de connaissances entre cancer et paludisme, deux syst\u00e8mes biologiques pr\u00e9sentant des r\u00e9gulations m\u00e9taboliques analogues.<br>Les travaux reposent sur des approches de mod\u00e9lisation mol\u00e9culaire et de chimie-informatique, combinant la conception <em>de novo<\/em> de nouvelles classes chimiques et l\u2019exploration de bases de donn\u00e9es pour identifier des fragments de mol\u00e9cules actives.<\/p>\n\n\n\n<p>Une seconde approche, \u00e0 l\u2019interface entre recherche th\u00e9rapeutique et bioproduction, consiste \u00e0 contr\u00f4ler le m\u00e9tabolisme \u00e9nerg\u00e9tique de la cellule. Le BIRL \u00e9tudie la bascule respiro-fermentaire \u2014 passage d\u2019un m\u00e9tabolisme respiratoire \u00e0 un m\u00e9tabolisme fermentaire \u2014 observ\u00e9e dans les cellules tumorales (<em>effet Warburg<\/em>) ou dans certains micro-organismes (<em>effet Crabtree<\/em>).<\/p>\n\n\n\n<p>L\u2019\u00e9quipe d\u00e9veloppe des mod\u00e8les formels de r\u00e9gulation m\u00e9tabolique pour rechercher des combinaisons th\u00e9rapeutiques capables d\u2019inverser l\u2019effet Warburg. Ce projet est conduit en collaboration avec le Prof. G. Bernot du laboratoire I3S (Universit\u00e9 C\u00f4te d\u2019Azur, Sophia Antipolis) et un doctorant, R. Khooderam.<\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column has-base-background-color has-background is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0;padding-right:var(--wp--preset--spacing--30);padding-bottom:0;padding-left:var(--wp--preset--spacing--30);flex-basis:33.33%\">\n<p class=\"has-tertiary-color has-text-color has-link-color has-large-font-size wp-elements-32b2fae2a45e2ba4c1239e25bda14bd9\"><strong>Qu\u2019est-ce que la bioinformatique ?<\/strong><\/p>\n\n\n\n<p class=\"has-tertiary-color has-text-color has-link-color wp-elements-1dc01978d326f1ea2bce74fdedc2fcb0\" style=\"border-style:none;border-width:0px\">La bioinformatique est une discipline \u00e0 l\u2019interface de la biologie, de l\u2019informatique et des math\u00e9matiques. Elle permet de mod\u00e9liser <em>in silico<\/em> des voies m\u00e9taboliques ou des processus cellulaires afin de mieux comprendre le vivant.<br>Elle repose sur l\u2019analyse de grands ensembles de donn\u00e9es issues notamment du s\u00e9quen\u00e7age g\u00e9nomique, de l\u2019imagerie biologique ou d\u2019exp\u00e9riences complexes.<br>S\u2019appuyant sur des mod\u00e8les math\u00e9matiques, logiques et statistiques, mais aussi sur les derni\u00e8res avanc\u00e9es de l\u2019intelligence artificielle, la bioinformatique ouvre la voie \u00e0 une compr\u00e9hension int\u00e9gr\u00e9e des syst\u00e8mes biologiques et \u00e0 la d\u00e9couverte de nouvelles solutions th\u00e9rapeutiques.<\/p>\n<\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-group has-global-padding is-layout-constrained wp-block-group-is-layout-constrained\">\n<div class=\"wp-block-columns is-style-header-columns is-layout-flex wp-container-core-columns-is-layout-28f84493 wp-block-columns-is-layout-flex\">\n<div class=\"wp-block-column is-vertically-aligned-top is-layout-flow wp-block-column-is-layout-flow\">\n<h5 class=\"wp-block-heading\" id=\"h-modelisation-des-transitions-metaboliques\"><strong>Mod\u00e9lisation des transitions m\u00e9taboliques<\/strong><\/h5>\n\n\n\n<p>Le contr\u00f4le du m\u00e9tabolisme cellulaire est au c\u0153ur de la recherche th\u00e9rapeutique et de la bio-production.<br>Le BIRL construit des mod\u00e8les formels du m\u00e9tabolisme \u00e9nerg\u00e9tique en s\u2019appuyant sur le formalisme de R. Thomas et sur les outils de v\u00e9rification de mod\u00e8les d\u00e9velopp\u00e9s par le Prof. Bernot (I3S). Ces approches permettent d\u2019\u00e9liminer les incoh\u00e9rences logiques et d\u2019identifier les combinaisons minimales d\u2019interventions n\u00e9cessaires pour induire un changement de ph\u00e9notype cellulaire.<br>Ce projet est r\u00e9alis\u00e9 en collaboration avec G. Bernot (I3S, Sophia Antipolis) et R. Koodheraam (Universit\u00e9 des Mascareignes, \u00eele Maurice).<\/p>\n\n\n\n<h5 class=\"wp-block-heading\" id=\"h-controle-et-analyse-des-bioprocedes\"><strong>Contr\u00f4le et analyse des bioproc\u00e9d\u00e9s<\/strong><\/h5>\n\n\n\n<p>Le second axe du laboratoire s\u2019int\u00e9resse \u00e0 la dynamique de croissance des populations cellulaires.<br>\u00c0 l\u2019\u00e9chelle d\u2019un bior\u00e9acteur ou d\u2019un tissu, le microenvironnement et la disponibilit\u00e9 des nutriments influencent directement la variabilit\u00e9 m\u00e9tabolique et la production de biomasse.<br>Le projet MAC-BIORE (Mod\u00e9lisation, Automatisme et Contr\u00f4le des Bior\u00e9acteurs) explore l\u2019influence de la diversit\u00e9 m\u00e9tabolique sur la productivit\u00e9 globale. Ces travaux sont men\u00e9s en collaboration avec le Prof. I. Boussaada du Laboratoire des Syst\u00e8mes Complexes de l\u2019IPSA, ainsi qu\u2019avec le L2S et le LGPM de CentraleSup\u00e9lec.<\/p>\n\n\n\n<p><strong>ANALYSES STATISTIQUES EN OENOLOGIE <\/strong><\/p>\n\n\n\n<p>Le BIRL \u00e9tudie \u00e9galement les processus de fermentation du mo\u00fbt de raisin en vin \u00e0 travers le comportement de la levure <em>Saccharomyces cerevisiae<\/em>, mod\u00e8le id\u00e9al pour comprendre la bascule respiro-fermentaire.<br>Ces recherches, men\u00e9es avec le Prof. R. Marchal (Universit\u00e9 de Reims Champagne-Ardenne) et la Dr. O. Arkoun (Laboratoire de Math\u00e9matiques Rapha\u00ebl Salem, Universit\u00e9 de Rouen-Normandie), visent \u00e0 d\u00e9velopper des m\u00e9thodes statistiques pour suivre la qualit\u00e9 des mo\u00fbts au cours du pressurage et discriminer les fractions pour un assemblage optimal.<\/p>\n\n\n\n<div class=\"wp-block-columns is-layout-flex wp-container-core-columns-is-layout-28f84493
