avec\nexploration par microscopie 3D<\/strong><\/p>\n\n\n\n<p>Les maladies neurod\u00e9g\u00e9n\u00e9ratives, qui\nreposent toutes sur des m\u00e9canismes de type prion (auto-propagation d\u2019agr\u00e9gats\nprot\u00e9iques pathog\u00e9niques), repr\u00e9sentent un risque soci\u00e9tal majeur avec une\naugmentation de leur pr\u00e9valence directement corr\u00e9l\u00e9e avec l\u2019augmentation de la\nlong\u00e9vit\u00e9 de la population mondiale et aucun traitement curatif ni aucun mod\u00e8le\nexp\u00e9rimental suffisamment pertinent.<\/p>\n\n\n\n<p>Ce travail utilise le potentiel des\ncellules reprogramm\u00e9es \u00e0 partir de patients adultes pour faire cro\u00eetre in vitro\ndes structures 3D autoassembl\u00e9es reproduisant des r\u00e9gions du cerveau humain\ncomme le cortex et obtenir un mod\u00e8le de \u201cmini-cerveau\u201d (organo\u00efdes c\u00e9r\u00e9braux).\nCe mod\u00e8le permet notamment d\u2019observer l\u2019apparition d\u2019agr\u00e9gats d\u2019A\u00df et de Tau\nphosphoryl\u00e9e, deux prot\u00e9ines qui s\u2019accumulent et se propagent de cellule en\ncellule dans la maladie d\u2019Alzheimer.<\/p>\n\n\n\n<p>Une telle approche, valid\u00e9e par une\nanalyse en microscopie 3D, pourra \u00e0 long terme s\u2019appliquer \u00e0 tout type\nd\u2019organo\u00efdes pour mod\u00e9liser l\u2019ensemble du corps humain
